巴马火麻仁木脂素酰胺类提取物的鉴定及清除自由基活性的研究

摘要:利用硅胶柱层析法以氯仿-甲醇(85:15)洗脱巴马火麻仁木脂素酰胺类的粗提物,经快原子轰击质谱(FAB-MS)核磁共振(NMR)鉴定为大麻酰胺A(cannabisin A)。以1,1-二苯基-2-苦基肼自由基(DPPH·)、超氧阴离子自由基(O2-)、羟自由基(OH·)三种不同自由基体系研究火麻仁木脂素酰胺类的粗提物、经大孔树脂纯化后的精提物及cannabisin A清除活性。结果表明在一定的剂量范围内,三者均有显著的活性清除作用,且与剂量成明显量效关系。精提物的清除作用最强,尤其是在OH·体系中,其清除OH·活性为粗提物的13倍。

Identification and free radical scavenging activity of lignanamides extract from Fructus Cannabis of Bama

Abstract: Cannabisin A was isolated from the crude lignanamides extract of Fructus Cannabis of Bama by chloroform-methanol(85:15) with silica gel column chromatography, and identified by FAB-MS, NMR.   Researching the scavenging activity of the crude lignanamides extract, the purified lignanamides extract by macroporous resin and cannabisin A by three diffierent free radical systems such as 1,1-Diphenyl-2-picryl- hydrazyl free radical(DPPH·), superoxide anion free radical(O2-), hydroxide free radical(OH·). The results showed that both of them had significant scavenging activity in three different free radical systems. Meanwhile, the scavenging capacity was dose-dependent in certain dose interval. The purified lignanamides extract had the strongest scavenging activity, especially in the OH·system, the activity of scavenging was 13 times compared with the crude lignanamides extract.

生物体内的能量代谢将氧气作为有氧代谢过程中的电子接受体,不可避免地产生活性氧(reactive oxygen species, ROS)自由基。相关研究已表明,自由基产生的氧化性损伤与衰老、肝病、人类免疫缺陷病毒(HIV)感染等均有密切关系[1-3]。因此,自由基清除剂逐步成为研究的热点。

巴马火麻仁源自于世界著名第五大长寿之乡——广西巴马,为桑科植物大麻Cannabis sativa L.的干燥成熟果实。目前,对火麻仁药理作用的研究主要是火麻仁油的抗氧化活性[4-5],而对其木脂素酰胺类(lignanamides of Fructus Cannabis, LFC)的药理活性研究较少,抗氧化活性在国内外均未见报道,因此本文通过研究LFC体外清除自由基活性,以期筛选能阻断氧自由基反应的天然、高效的抗氧化活性物质,为火麻仁进一步的开发利用提供可靠的科学依据。
